Title: EZH2 may methylate SRSF1 and affect its function in CML

Abstract:
EZH2 is the enzymatic component of the PRC2 complex which plays an important role in Histone modification and gene regulation. However, we and others have shown that EZH2 can also methylate non-histone proteins as well as interacts with other proteins and RNA leading to alternate functions and effect on gene expression. In an attempt to understand these non-canonical functions, we performed proteomics study to identify interacting partners with EZH2. In our proteomics study, EZH2 interacts with a large number of splicing factor. We validated that SRSF1 is one of the splicing factor that can be methylated by EZH2. SRSF1 has oncogenic function in CML and this is mediated partly by methylation induced by EZH2.
